Title: "Genetic influence in cognitive development: DNA estimates"
Abstract: It is now possible to use DNA to estimate genetic influence in any large sample of unrelated individuals (Genome-wide Complex Trait Analysis, GCTA). Our work using GCTA has confirmed three findings about cognitive development that previously emerged from the classic twin design: (1) Diverse cognitive abilities are all moderately heritable; (2) The same genes affect diverse cognitive and learning abilities; and (3) From childhood to adolescence, the heritability of general cognitive ability (aka intelligence) increases even though genetic effects are stable. These GCTA results suggest that genetic influences on cognitive abilities, and presumably the brain structures and functions that underlie them, can be substantially accounted for by the additive effects of common variants used on current DNA arrays. The results also imply that genetic effects are highly polygenic (many genes affect each trait) and highly pleiotropic (each trait is affected by many genes).

Abstract: Water is a profoundly unusual liquid, and its peculiarities may make it uniquely suited to act as life's matrix. Even if this were not so, however, we should expect the effects of nanometre-scale confinement and inhomogeneities owing to surface effects to alter the liquid's properties in the cell relative to those in the bulk. Whether water's unusually high degree of local structure makes such influences even more marked than for 'normal' liquids remains an open question, with potentially important consequences for biomolecular interactions.

Abstract: The tumour suppressor gene p53 is a well-known transcriptional activator with targets ranging from cell cycle checkpoint, apoptosis, repair and senescence, all designed to regulate the cell’s response to stress.
Recent studies have shown that expression and mitochondrial localisation of p53 increase after neonatal HI, as do a number of p53-regulated pro-apoptotic targets; we therefore hypothesised that inhibition of p53 translocation and its subsequent downstream interactions may be protective in neonatal hypoxia-ischaemia (HI)-mediated stress.
In a recent study, our collaborators found that inhibition of mitochondrial p53 resulted in long-lasting protection in a mouse model of HI. Injection of PFT-μ (Pifithrin-µ; inhibitor of mitochondrial p53) post-HI in rats and mice resulted in reduced loss of grey and white matter and prevention of behavioural deficits seen in the vehicle-treated animals.
Our subsequent in vitro studies (and the focus of this talk) have generated a more detailed knowledge of the regulation of p53 by pifithrin, and have implicated an unexpected mechanism contributing to this protection.

Abstract: Looking for a friend in the crowd or avoiding a sudden danger are two actions based on the quality of our visuo-spatial attention. Behavioural tests and brain imaging technology demonstrated that visuo-spatial attention is a specialized function of the right hemisphere of the brain. However, anatomical features supporting this specialization have remained elusive. We scanned volunteers with a novel brain imaging technology that can depict the white matter connections, and we measured the degree of specialization of the right hemisphere for visuo-spatial attention with behavioural tests. Exploring the brain circuits, we found white matter connections whose size predicts the degree of specialization of the right hemisphere for visuo-spatial attention. Whilst a damage of this connection in the right hemisphere can predict chronicity of the visuospatial deficits, the lateralisation of this same connection may be predictive of visuo-spatial recovery.
